Preparing a Transverse Section of a Stem

Using a carrot block and a very sharp razor blade cut extremely thin sections of the stem and root and float the sections in water in your beaker.

Mount the thinnest two sections onto slides (they look the most transparent in water)

Stain them with a drop of iodine and add a cover slip.

SECRETS of a GOOD SECTION:

·         use a newly sharpened blade 

·         keep the blade and the carrot moist

·         use a sawing motion across the leaf as you cut

·         use a small brush to transfer your sections to slides

 Drawing cells of the stem or root at low power x100

Adjust the slide and microscope to get a good view of the stem or root.  

Identify the different types of cell by looking at the thickness of the cell wall and its shape or size.

In microscope drawings of tissue distribution you only need to;

Draw an outline to show the position of each of the six types of tissue:

Make sure that it is accurate in shape, size and position.
